FRIPS:
Free radical-induced peptide sequencing
The Technique
The FRIPS technique
was developed in the Beauchamp group for the selective sequencing of
peptides. A free radical initiator, Vazo 68 from DuPont, is
coupled to the N-terminus of the peptide in question. Collisional
activation results in loss of a part of the radical initiator, leaving
a carbon centered radical. A subsequent stage of collisional
activation leaves an acetyl radical attached to the N-terminus of the
peptide. It is this radical moiety which reacts with the peptide
backbone and sidechains to fragment it.
